If you take one hit you still have oxygen in your lungs, so you live a while longer. If you take a second hit, you have depleted the oxygen in your lungs. At some point you pass out from lack of oxygen. That is kinda rare because people usually only take one hit at a time. But inhaling nitrogen is different: you can pass out with a single breath.
